10x chromium

10x Chromium is a platform for profiling RNA at the single-cell level, with the capability for multiomic analysis, such as protein and TCR/BCR.

Some of the available assays include Universal Gene Expression, which allows for various multiomic readouts from the same cell (various species).

profiling using gel beads and unique barcoded probes

The Chromium Single-Cell platform uses Gel Beads, which are coated with assay-specific barcoded probes to capture the molecules of interest.

The basic workflow involves loading a suspension of single cells mixed with assay-specific reagents, Gel Beads, and enzymes onto a microfluidic chip. The Chromium instrument then moves the cells through the chip channels to produce tens of thousands of GEMs. The Gel Beads are dissolved within each GEM droplet, and molecules of interest from each cell are captured and barcoded. Once barcoded, a common 10x barcode is shared for all fragments from the same cell. These barcoded fragments are then pooled to create sequencing libraries.
